Is the nozzle lifting before it makes a travel move?

You might also check the wiring of all three steppers to make sure a pin isn't crawling out of the connector shell.

You should really only need to run the First Probe and Bed Mapping macros to fully calibrate your machine.

If the hot-end fan is running, it can cause this. It shouldn't affect how the z-probe operates when doing calibration.

400mm/sec is only valid for Duet-equipped machines. Dial it back to around 200mm/sec.

What I would do is try the HE280 profile for Cura. You can get all the pre-built Cura profiles here: https://github.com/seemecnc/slicing-pro ... master.zip. The HE280 is very similar in performance to the E3Dv6, so it should work with no tweaking.

FYI, if you're not doing a z-lift on retract (and thus travel), the nozzle could be striking the print and causing the layer shift. I seriously doubt it's the weight of the effector platform. Your travel speed could also be a factor - anything over around 220mm/sec on a RAMBo can be problematic.

The only version of the firmware that you should be running is the one on SeeMeCNC's git repo. It has customizations for the HE280 hot end that the main branch of Repetier Firmware does not have.

Are you printing from USB? If so, is your computer going to sleep?

Also, make sure that if you're compiling/uploading firmware, you're using the 1.6.0 version of the Arduino IDE.

Did you delete config-override.g? You need to do that and then re-run first calibration. (If you created a bed map, delete and re-create that as well.)

muresearch, it sounds to me like the z-zero of the machine isn't set correctly. What happens when you go through the calibration process via the front panel of the printer? You need to make sure that you compiled the firmware with version 1.6.0 of the Arduino IDE. Newer versions produce a very flaky...
